How to activate Multisite blog with WP 3.0

1. Hello Guys,
   I have recently installed a new WP 3.0.1 installation and trying to set it as WP mu site.
   But I am not getting where to change or update settings.
   Any helpful link or steps to solve this issue would be a great help.
