I am asking one of my team members to recheck it and assit you in the day.
Is there any staging site where we can disable some other plugins too just to find the conflict. Or is this site still in development and not live? In that case, we can disable some of the plugins here if you allow, to find the conflict.

That does not seem to be related to that issue.
Also, the BuddyPress change swill affect if you haven’t overridden the buddypress.js file.
Try to check with your theme devloper(or temporarily rename the file) to see if it related to your custom file or not.

MediaPress does not have any upper or lower limit on the file size. It is the server that applies the limits.
Please see the previous replies or contact hosting support for troubleshooting.
